基于g-C3N4纳米片、铜离子和邻苯二胺构建的比率荧光探针检测辅酶A

摘要: 开发了一种基于g-C3N4纳米片、铜离子(Cu2+)和邻苯二胺(OPD)的比率荧光探针,用于检测辅酶A (Coenzyme A,CoA)。分别采用g-C3N4纳米片和Cu2+氧化OPD后的产物2,3-二氨基吩嗪(oxOPD)作为荧光指示剂,Cu2+和oxOPD作为淬灭剂,检测体系中g-C3N4纳米片的荧光被淬灭,oxOPD荧光增强。CoA与Cu2+形成络合物,减少了体系中作为氧化剂和淬灭剂的游离Cu2+的含量,导致OPD的氧化过程受到抑制,oxOPD的含量减少,荧光强度降低,g-C3N4纳米片的荧光恢复。以g-C3N4纳米片的荧光发射强度与oxOPD的荧光发射强度比(F440/F560)作为响应信号,用于比率荧光检测。在最佳实验条件下,采用本方法检测CoA,在1.0~100.0 μmol/L浓度范围内具有良好的比率荧光响应,线性方程为F440/F560=0.00502CCoA(μmol/L)+0.1079,R2=0.984,检出限(LOD,3 S/m)为0.6 μmol/L。此比率荧光检测体系具有良好的选择性,人血清中CoA的加标回收实验结果表明,此体系在复杂生物基质检测中具有良好的应用潜力。
