多酚氧化酶在聚乙烯醇接枝谷氨酸枝脂Cu(Ⅱ)配合物上的固定化
摘要:
研究了固定化酶载体——聚乙烯醇接枝谷氨酸树脂的合成及树脂络合金属铜离子后对多酚氧化酶的固定化.用红外光谱表征了树脂的结构,用DTA测定了树脂的热行为.考察了固定化酶的特性.结果表明,该树脂与Cu2+配位后的高分子配合物能较好地对多酚氧化酶固定化,此固定化酶最适pH值为5.59和6.64,能重复多次使用,经5次使用后其活力仍保留20%.固定化酶的热稳定性比游离酶高,而米氏常数基本相同.

Immobilization of Polyphenol Oxidase by Coordination with Cu(Ⅱ) in Polyvinyl Alcohol Grafted Glutamic Acid Resin
Abstract:
The synthesis of the immobilization carrier of polyphenol oxidase(PPO): polyvinyl alcohol grafted glutamic acid resin(PGAR) has been studied and the structure of the PGAR has been characterized by IR and DTA techniques. The results showed that the PPO was immobilized onto the carrier resin through coordination with Cu2+, forming PGAR-Cu-PPO. The optimum pH for immobilization was 5.59 and 6.64, and the Michaelis-Menten constant was of 1.70×10-2 mol/L, both are the same as those for free PPO. The immobilized PPO can be used repeatedly and the activity was found reduced by 80% after being reused 5 times.
