Abstract:
The fluorescence spectra and photodimerization of naphthalene end-labelled poly(ethylene glycol)oligomers(N-Pn-N)in methanol in the prescence of alkali cations have been investigated. The ion enhances or inhibits intramolecular excimer formation and photodimerization of N-Pn-N,depending on the size of the ion and the length of the polyether chain. This observation suggests that the polyether chain binds the ion, thus adopts a crown ether-like conformation.

[Abstract](453) [FullText HTML] [PDF 142KB](0)
Abstract:
Melittin is a small basic peptide which has a high affinity for calmodulin(CaM).In a previous paper, the authors have demonstrated that the deletion peptide Mel(12~25),that is,Mel15 has almost the same binding activity as the intact native peptide(1).In this study, three analogues of Mel15 were synthesized, the role of amphiphilic α-helix of peptides on their binding to CaM were studied.According to the calculation of hydrophobic moment and the analysis of the secondary structure,we proposed that the role of amphiphilic structure is not absolute, since the dissociation constants for the binding of different analogs to CaM bear no predictable relation to the calculated hydrophobic moment and the average hydrophobicity

Abstract:
We report here a fast staining method for single-stranded DNA fixed on nitrocellulose membrane using M-meso-tetra(3-N-methylpyridyl)-porphyrin(M-TMPyP,M:Mn3+,Fe3+,Pt+).The optimal staining solution consists of 54~108μg/ml M-TMPyP in 25~200 mmol/L phosphate buffer solution(pH 5.0~8.0).When DNA dot on NC membrane is treated with the staining solution at room temperature for 1~2 minutes with continuous stirring,it turns to brown-yellow.10pg per dot of single-stranded DNA is visible. It is a useful tool to study the DNA blotting, and for recognition of the special bands of DNA in a complete electrophoretogram of DNA blot. The staining mechanism is also discussed.

[Abstract](448) [FullText HTML] [PDF 102KB](0)
Abstract:
The mathematical equations of the effective percentage of a complexant and its complex-ratio with a metal have been developed by means of the β-correction pinciple. From a real abeorbance of the chelate produced, its true abeorptivity can be determined accurately. The β-correction spectrophotometry was studied for the determinations of the desired parameters above, using the reaction of lead with Eriochrome Black T(EBT)at pH 8.2.The results show that the analytical ansitivity and precision by β-correction method are both much higher than those by the single wavelength method, the complex-ratio of Pb(Ⅱ)with EBT equals to 1:4 and the true molar absorptivity of Pb(EBT)4chelate at 550 nm is 3.53×104L.mol-1.cm-1.

Abstract:
It was found that ribonucleic acid(RNA)compiexes with Tb(Ⅲ)at pH=5.0-6.5,which then emits strong characterlstic fluorescence of Tb(Ⅲ).The excitation and emission wavelengths are 288nm,494nm and 545nm,respectively.A linear relationship is obtained between the fluorescence intensity and RNA concentration in the range of 0.1 ug/ml-10 ug/ml.The detection limit is 8.0×10-10mol/L.This method can be used to determine RNA in the presence of adenylic acid, uridylic acid and cytidylic aci.
