靶向超高效液相色谱-串联质谱联用方法同时测定结直肠癌患者血浆中32种氨基酸含量

摘要: 建立了靶向超高效液相色谱-串联质谱联用方法(UHPLC-MS/MS)用于同时测定结直肠癌患者血浆中32种氨基酸含量。采用Agilent Zorbax SB-C18色谱柱(150 mm×3.0 mm,5 μm)、二元梯度洗脱系统(流动相A为0.02%七氟丁酸-0.2%甲酸溶液,流动相B为甲醇)进行色谱分离,运行时间为10 min。采用蛋白沉淀法进行样品前处理,选择多反应监测模式,以电喷雾电离正离子模式获取数据。32种氨基酸和3种内标氨基酸的提取回收率为50.4%~182.4%,基质效应为41.0%~136.4%;32种氨基酸在其相应线性范围内的线性相关系数均大于0.99,日内准确度和日间准确度分别为-14.3%~13.7%(RSD≤8.8%)和-14.4%~13.4%(RSD≤12.2%),不同条件下的稳定性偏差均在±15%之内。应用本方法同时测定了6名结直肠癌患者血浆中32种氨基酸含量。
